The insert contains the following sites separated by (kb) (approximate): SphI- 1.4- SacI- 0.6- HindIII- 1.6- KpnI- 0.6- HindIII- 0.8- SphI- 1.2- SphI. Restriction digests of the clone give the following sizes (kb): EcoRI/SalI--3.75, 3.5, 3.3, 2.9, 2.3, 2.0, 0.94; XhoI--18.7; XbaI--14.6, 4.2; XbaI/AvaI--4.4, 3.5, 3.1, 2.3, 2.0, 1.8, 1.5; BamHI--18.7. The smallest subclone capable of complementing msf1-1, msf1-3, and msf1-4 mutations contained a 2.2 kb SacI/KpnI fragment. MSF1 is a nuclear gene encoding the alpha subunit of the mitochondrial phenylalanyl-tRNA synthetase. The probe, pG120/T1, has been deposited in Escherichia coli RR1 as ATCC 37664 and in Saccharomyces cerevisiae C303/L2 as ATCC 66317. |
