Restriction digests of the clone give the following sizes (kb): HindIII--4.4, 1.2; SalI--4.8, 0.7; XbaI--uncut. To induce expression of the insert, grow in LB with limiting concentrations of tryptophan. A genomic NruI/SmaI fragment was modified and cloned into pTrp3. Contrary to the description published in J. Bacteriology 173, 6849-6858, 1991, this construct appears to have had HindIII sites engineered up and downstream of the coding region. |
