Restriction digests of the clone give the following sizes (kb): BamHI--23.0, 16.0, 2.4; HindIII--27.5, 4.4, 2.9, 2.6, 2.1, 1.7, 1.1, 0.82; SacI--24.0, 10.0, 9.8. The complete coding sequence can also be isolated from partial digests of the clone as a 6.7 kb HindIII-BamHI fragment or a 4.7 kb HindIII-SmaI fragment. Insert contains the following restriction sites approximate kb from the 5' end): BamHI--18.1; BglI--13.3, 14.5; EcoRI--16.9; HindIII--14.0, 14.8, 19.3; PstI--14.3, 14.6; SacI--10.2; SalI--12.2, 12.8; SmaI--16.1; XhoI--15.3, 20.1. Insert contains two SacI fragments of 10.2 kb each. The downstream fragment encodes the six open reading frames involved in the expression of toluene-4-monooxygenase. |
